Course objectives:
|
The aim of this course is to make students familiar with principes of safe work in laboratory, methods of sampling, preparation and conservation of plat and animal materials, to introduce students to use of main laboratory instruments (microscopes, binocular lens) and to practice of laboratory work (some basic laboratory methods used in biology).

Requirements on student
|
The student must be active in laboratory courses. He has obligation to make the protocols and accomplish the the credit test. He must master basic laboratory skills.

Content
|
1. Labor safety
2. History of microscopic techniques
3. Basic optical principles
4. Construction of microscope, preparation of preparations.
5. Working with a stereoscopic microscope.
6. Electron microscopy, preparation of electron microscopy specimens
7. Pipetting, DNA isolation, agar gel preparation and DNA visualization on the gel, preparation of bacterial cultures.
8. Collection, preparation and preservation of plant and animal material
9. Types of staining of microscopic preparations
10. Watering of the tissue into paraffin binders, cutting and preparation of preparations
11. Documentation of biological objects (digital photography, drawing and computer graphics).
12. Basic laboratory and measuring instruments in biology (pH meter, oximeter, fluorimeter, electrophoresis, centrifuge, etc.).
13. Credit test.

Knowledge - students are expected to possess the following knowledge before the course commences to finish it successfully: |
know labor safety in the laboratory |
be able to distinguish laboratory equipment and aids |
know ways of collecting, preparing and preserving plant and animal material |
know the principles of light and electron microscopy |
to be able to prepare microscopic preparations and be able to work with a stereoscopic microscope |
know some basic laboratory methods used in biology and be able to document biological objects (classic and digital photography, drawing and computer graphics) |

to be able to work with a light microscope and a stereomicroscope |
on the basis of the assigned task to specify the goals, results and conclusions of their own observation at the gymnasium level |
